Systematic Gene Tagging with CRISPR/Cas9 in hiPSC’s to Illuminate Cell Organization and Dynamics across Cell States
ABSTRACT: The Allen Institute for Cell Science  is creating a dynamic visual model of hiPSC organization by generating a collection of fluorescent-tagged clonal hiPSC lines. Our approach uses CRISPR/Cas9 to fluorescently tag proteins that localize to the major organelles of the cell. Live cell imaging, image analysis, modeling and visualization, and open distribution to the scientific community define our endeavor. In experiments initiated to date we have generated 25 hiPSC lines with fluorescent tags labeling major cellular structures with key genetic, cell biological, and stem cell validation. We will demonstrate the utility of these cell lines for generating image-based integrated models of cell organization and dynamics in stem cells and iPSC-derived cardiomyocytes. We will also discuss the integration of genomic data with live cell imaging to explore cell states and transitions using this collection of endogenously tagged stem cell lines.
